ThepiwigeneofDrosophilabelongstoanovelclassofevolutionarilyconservedgenes(thepiwiorArgonautefamily).ThePiwifamily‘isrequiredforgerm-andstem-celldevelopmentininvertebrates,andtwoPiwimembers,MIWIandMILI(Miwilike;Piwil2;Piwi-likeprotein2),areessentialforspermatogenesisinmouse.MIWIandMILIpartnerwithPiwi-interactingRNAs(piRNAs)whichare26–31nucleotides(nt)inlength—clearlydistinctfromthe21–23ntofμRNAs(miRNAs)orshortinterferingRNAs(siRNAs).AsearchformurinesmallRNAsthatmightprogramPiwiproteinsfortransposonsuppressionrevealeddevelopmentallyregulatedpiRNAloci,someofwhichresembletransposonmastercontrollociofDrosophila.MILImightbeprogrammedbydistinctpiRNApopulationsatdifferentstagesofgermcelldevelopment,suggestingMILIplaysanimportantroleinmeioticprophaseprogression.Playsacentralroleduringspermatogenesisbyrepressingtransposableelementsandpreventtheirmobilization,whichisessentialforthegermlineintegrity.Playsanessentialroleinmeioticdifferentiationofspermatocytes,germcelldifferentiationandinself-renewalofspermatogonialstemcells.Itspresenceinoocytessuggeststhatitmayparticipatetosimilarfunctionsduringoogenesisinfemales.ActsviathepiRNAmetabolicprocess,whichmediatestherepressionoftransposableelementsduringmeiosisbyformingcomplexescomposedofpiRNAsandPiwiproteinsandgovernthemethylationandsubsequentrepressionoftransposons.DirectlybindspiRNAs,aclassof24to30nucleotideRNAsthataregeneratedbyaDicer-independentmechanismandareprimarilyderivedfromtransposonsandotherrepeatedsequenceelements.AssociateswithprimarypiRNAsinthecytoplasmandisrequiredforPIWIL4/MIWI2nuclearlocalizationandassociationwithsecondarypiRNAsantisense.ThepiRNAprocessactsupstreamofknownmediatorsofDNAmethylation.ParticipatestoapiRNAamplificationloop.Besidestheirfunctionintransposableelementsrepression,piRNAsareprobablyinvolvedinotherprocessesduringmeiosissuchastranslationregulation.IndirectlymodulateexpressionofgenessuchasPDGFRB,SLC2A1,ITGA6,GJA7,THY1,CD9andSTRA8.Inhibitstumorcellgrowthwhenrepressed.Whenoverexpressed,actsasanoncogenebyinhibitionofapoptosisandpromotionofproliferationintumors.
