Peroxiredoxin(Prx)isanantioxidantenzymedetoxifyingreactiveoxygenspeciesandhasacysteineattheiractivesite.Prxenzymesmodulatevariousreceptorsignalingpathwaysandprotectcellsfromoxidativelyinduceddeath.PrxItoIVhavetwoconservedCysresiduescorrespondingtoCys51andCys172ofmammalianPrxI.Theactivesitecysteine(Cys51)isoxidizedtocysteinesulfenicacid(Cys51-SOH)whenaperoxideisreduced.BecauseCys51-SOHisunstable,itformsadisulfidewithCys172-SHwhichcomesfromothersubunitofthehomodimer.ThedisulfideisthenreducedbacktothePrxactivethiolformbythethioredoxin-thioredoxinreductasesystem.However,theformationofthedisulfideisaslowprocess.Thusunderoxidativestresscondition,thesulfenicintermediate(Cys51-SOH)canbeeasilyoveroxidizedtocysteinesulfinicacid(Cys-SO2H)orcysteinesulfonicacid(Cys-SO3H)beforeitisabletoformadisulfide.RecentstudiessuggestthatoveroxidizedPrxcanbereducedbacktotheactiveformduringrecoveryafteroxidativestress.Anti-Prx-SO3antibodyrecognizesbothsulfinicandsulfonicformsofPrxanddetectsoveroxidizedPrxenzymesinH2O2-treatedcellswithhighsensitivityandspecificity.Involvedinredoxregulationofthecell.Reducesperoxideswithreducingequivalentsprovidedthroughthethioredoxinsystembutnotfromglutaredoxin.Mayplayanimportantroleineliminatingperoxidesgeneratedduringmetabolism.Mightparticipateinthesignalingcascadesofgrowthfactorsandtumornecrosisfactor-αbyregulatingtheintracellularconcentrationsofH2O2.ReducesanintramoleculardisulfidebondinGDPD5thatgatestheabilitytoGDPD5todrivepostmitoticmotorneurondifferentiation.
