PeroxiredoxinVI(Prx6,1-CysPrx)isamemberofPeroxiredoxinFamily,anantioxidantenzymethatdetoxifiesreactiveoxygenspeciesandhasacysteineattheiractivesite.Sixisoforms(Prx1to6)ofPrxexistinalleukaryoticcells.Theseisoformsareclassifiedintothreesubgroups(2-Cys,atypical2-Cys,and1-Cys).Prx6modulatesvariousreceptor-signalingpathwaysandprotectscellsfromcelldeathinducedbyoxidativestress.Theactivesitecysteine(Cys47)isoxidizedtocysteinesulfenicacid(Cys47-SOH)byH2O2.However,theresultingCys47-SOHdoesnotformadisulfidebondbecauseofunavailabilityofanotherCys-SHnearby.ItcanbereducedbynonphysiologicalthiolssuchasDDTbutisnottransformedbyThioredoxin/ThioredoxinReductaseorGSH.Occasionally,thesulfenicintermediateishyperoxidizedtosulfinicorsulfonicacid,resultingininactivationofperoxidaseactivity.Involvedinredoxregulationofthecell.CanreduceH2O2andshortchainorganic,fattyacid,andphospholipidhydroperoxides.Mayplayaroleintheregulationofphospholipidturnoveraswellasinprotectionagainstoxidativeinjury.
