Periostinisa90-kDamatricellularproteinthatconsistsofatypicalsignalsequence,followedbyacysteine-richregion,anEMIdomain(protein-proteininteractions),fourtandemfasciclin-likedomainsthatareresponsIBLeforintegrinbinding,andaC-terminalregion.Periostinwasoriginallyisolatedasanosteoblast-specificfactorthatfunctionsasacelladhesionmoleculeforpre-osteoblastsandinosteoblastrecruitment,attachmentandspreADIng.PeriostinisalsoinvolvedinmanyfundamentalBIOLOGicalprocessessuchascellproliferation,cellinvasionandangiogenesis.Periostinexpressionisincreasedbybothtransforminggrowthfactorβ1(TGF-β1)andbonemorphogeneticprotein2(BMP-2).Changesinperiostinexpressionarecommonlydetectedinvariouscancersandpre-cancerousconditions,andperiostinmaybeinvolvedinregulatingcancercellactivitiesthatcontributetotumOrigenesis,cancerprogressionandmetastasis.Periostinhasshowntobeinvolvedinmanyaspectsofallergicinflammation,suchaseosinophilrecruitment,airwayremodeling,developmentofaTh2phenotypeandincreasedexpressionofinflammatorymediators.ItisevaluatedasabioMarkerforbronchialasthmaandairwayinflammation.
