Platelet-derivedgrowthfactors(PDGFs)havebeenimplicatedinthecontrolofcellproliferation,survivalandmigration.ThePDGFfamilyofgrowthfactorsconsistsoffivedifferentdisulphide-linkeddimersbuiltupoffourdifferentpolypeptidechainsencodedbyfourdifferentgenes.Thesesisoforms,PDGF-AA,PDGF-AB,PDGF-BB,PDGF-CCandPDGF-DD,actviatworeceptortyrosinekinases,PDGFreceptorsαandβ.Thusfar,gene-targetingexperimentshavebeenattemptedtocreateknockoutmicedeficientforPDGFR-αorPDGFR-β.Thosemice,however,diedeitherattheembryonicstageorseveraldaysafterbirth.Platelet-derivedgrowthfactorreceptors,PDGFR-αandPDGFR-β,havefiveextracellularimmunoglobulin-likedomainsandanintracellulartyrosinekinasedomain.UponbindingaPDGF,thereceptorsformhomo-andheterodimers.Dimerizationofthereceptorsjuxtaposestheintracellularpartofthereceptors,whichallowphosphorylationintransbetweenthetworeceptorsinthecomplex.Theseautophosphorylationprovidedockingsitesfordownstreamsignaltransductionmolecules.Morethan10differentSH2–domain-containingmoleculeshavebeenshowntobindtodifferentautophosphorylationsitesinthePDGFα-andβ-receptors.Therearesignaltransductionmoleculeswithenzymaticactivity,suchasPI3-kinase,PLC-γ,Src,SHP-2,GAP,aswellasadaptormoleculessuchasGrb2,Shc,Nck,Grb7andCrk,andStats.Eachofthedifferentpartnersrecruitedbytheactivatedreceptorinitiatesdifferentsignalingpathways,makingpossIBLeagreatvarietyofcellularresponse.ReceptorthatbindsspecificallytoPDGFBandPDGFDandhasatyrosine-proteinkinaseactivity.PhosphorylatesTyrresiduesattheC-terminusofPTPN11creatingabindingsitefortheSH2domainofGRB2.
