Synucleinisasmall,solubleproteinprimarilyexpressedinneuraltissue.Thesynucleinfamilyincludesthreeknownproteins:α-synuclein,β-synuclein,andγ-synuclein.α-Synucleinisaveryconserved,smallacidicprotein(140residues)withamolecularweightofapproximately19kDathataccountsforabout1%oftotalproteininneurons.ItisconcentratedprimarilyinpresynapticaxonterminalsandplaysacentralroleininitiatingParkinson’sdisease.ItistheprincipalconstituentofLewybodieswhichareformedbyproteinaggregationandinclusionbodyformation.α-Synucleinlackssecondaryortertiarystructure,soitbelongstothefamilyofnativelyunfoldedproteins,manyofwhichactaschaperones.Muchofthisα-synucleinhasbeenpost-translationallymodifiedtoincreasetherateofoligomerization.Mutantformsofα-SynucleinareassociatedwithautosomaldominantParkinson’sdisease.Themutationsincreasetheaggregationrateoftheresultantα-synucleinβ-Synucleinisfoundprimarilyinbraintissueandisseenmainlyinpresynapticterminals.Itispredominantlyexpressedintheneocortex,hippocampus,striatum,thalamus,andcerebellum.β-Synucleinmayprotectthecentralnervoussystemfromtheneurotoxiceffectsofα-synucleinbyinhibitingaggregationofα-synucleinwhichoccursinneurodegenerativediseasessuchasParkinson"sdisease.γ-Synucleinisasynucleinproteinfoundprimarilyintheperipheralnervoussystem(inprimarysensoryneurons,sympatheticneurons,andmotorneurons)andretina.γ-SynucleinexpressioninbreasttumorsisaMarkerfortumorprogression.Non-amyloidcomponentofsenileplaquesfoundinAlzheimerdisease.CouldactasaregulatorofSNCAaggregationprocess.Protectsneuronsfromstaurosporineand6-hydroxydopamine(6OHDA)-stimulatedcaspaseactivationinaTP53/p53-dependentmanner.ContributestorestoretheSNCAanti-apoptoticfunctionabolishedby6OHDA.NotfoundintheLewybodiesassociatedwithParkinsondisease.
