p38MAPKcascaderegulatesavarietyofcellularresponsestostress,inflammationandothersignals.p38MAPKisrelativelyinactiveinthenon-phosphorylatedformandbecomesrapidlyactivatedbydualphosphorylationofaThr-Gly-Tyrmotifs.Therearefourisoformsofp38MAPK,whichdifferintheirtissueexpressionandaffinityforupstreamactivatorsanddownstreameffectors.Whencellsareexposedtotumornecrosisfactor-α,interleukin-1,heatshock,orotheractivatingstimuli,activationofMAPKkinase-3and–6occursbyphosphorylation.ActivatedMAPKkinase-3/6phosphorylateeachresidueofThr180andTyr182inp38MAPK.Phospho-p38MAPKactivatesATF-2,CHOP-1,MEF-2andothertranscriptionfactorsthroughphosphorylation.Respondstoactivationbyenvironmentalstress,pro-inflammatorycytokinesandlipopolysaccharide(LPS)byphosphorylatinganumberoftranscriptionfactors,suchasELK1andATF2andseveraldownstreamkinases,suchasMAPKAPK2andMAPKAPK5.Playsacriticalroleintheproductionofsomecytokines,forexampleIL-6.MayplayaroleinstabilizationofEPOmRNAduringhypoxicstress.IsoformMxi2activationisstimulatedbymitogensandoxidativestressandonlypoorlyphosphorylatesELK1andATF2.IsoformExipmayplayaroleintheearlyonsetofapoptosis.
