p21,alsoknownascyclin-dependentkinaseinhibitor1AorCDKN1A,directlyinhibitstheactivityofcyclin-cdk2andcyclin-cdk4complexes.Itmainlyinhibitstheactivityofcyclin/cdk2complexesandnegativelymodulatescellcycleprogression.p21alsoplaysotherfundamentalrolesincludingtranscriptionalregulationandthemodulationofapoptosis.p21formspartofafamilyofstructurallyrelatedproteinscomprisingp21,p27andp57thatallsharetheabilitytoinhibitawiderangeofcyclin-dependentkinaseholoenzymes.Besidesitscdkinhibitorfunction,p21isrecognizedtoplayawidevarietyofphysiologicalroles,manyofwhichrelyonitsnuclearlocalization.p21canbindtoproliferatingcellnuclearantigen(PCNA)therebyblockingDNAsynthesis.p21functionsasaregulatorofcellcycleprogressionatSphase.Theexpressionofp21iscontrolledbythetumorsuppressorproteinp53.Eachofthesefunctionsofp21isachievedthroughdirectp21/proteininteractionsandthesubcellularlocalizationofp21playsanimportantpartindictatingthebindingpartnerstowhichp21isexposed.Maybetheimportantintermediatebywhichp53mediatesitsroleasaninhibitorofcellularproliferationinresponsetoDNAdamage.Bindstoandinhibitscyclin-dependentkinaseactivity,preventingphosphorylationofcriticalcyclin-dependentkinasesubstratesandblockingcellcycleprogression.
