ThethreeRb(retinoblastoma)familyproteins,p107,p130,andpRB,arealsoknownas"pocketproteins",aftertheirconservedpocketregion.Membersofthefamilyformpartofasignal-transductionpathwaycalledtheRbpathway,whichisimportantincell-cycleregulationandhaverolesingrowthsuppression,differentiationandapoptosis.ThepocketdomainisresponsIBLeforinteractionwithtranscriptionfactors,cyclins,andcyclin-dependentkinases(CDKs).TheyplaycrucialrolesinthecellcyclethroughinteractionwithmembersoftheE2Ftranscriptionfactorsfamily.TheyareactingonthecellcyclebetweenG0andSphases,primarilythroughbindingandinactivationoftranscriptionfactors.Thethreepocketproteinsconsistofanamino-terminaldomain,apocketregioncomposedoftwoconserveddomains(AandB)separatedbyaspacerregion,andacarboxy-terminaldomain.ThehumanRBgene(whichencodesaproteinof105kDaandisalsocalledp105)wasfirstidentifiedwhenbothfamilialandsporADIcretinoblastoma,aformofmalignanttumoroftheretina.Theretinoblastomaproteinwasoriginallydescribedasatumorsuppressor,asitwasfoundtobemutatedinmanyformsofcancer.Keyregulatorofentryintocelldivision.Directlyinvolvedinheterochromatinformationbymaintainingoverallchromatinstructureand,inparticular,thatofconstitutiveheterochromatinbystabilizinghistonemethylation.RecruitsandtargetshistonemethyltransferasesSUV420H1andSUV420H2,leadingtoepigenetictranscriptionalrepression.ControlshistoneH4"Lys-20"trimethylation.Probablyactsasatranscriptionrepressorbyrecruitingchromatin-modifyingenzymestopromoters.PotentinhibitorofE2F-mediatedtrans-activation,associatespreferentiallywithE2F5.BindstocyclinsAandE.BindstoandmaybeinvolvedinthetransformingcapacityoftheadenovirusE1Aprotein.Mayactasatumorsuppressor.
