NCK1isoneoftheadaptorproteinswhichmediatespecificprotein-proteininteractionsinsignalingprocesses.AdaptorproteinsusuallycontainseveraldomainslikeSH2(Srchomology2)andSH3whichallowspecificinteractionswithotherspecificproteins.NCK1andNCK2showinghighsequenceidentity(68%)havethreeSH3domainsandaC-terminalSH2domain.BothofthembindreceptortyrosinekinasessuchasPDGFRandothertyrosinephosphorylatedproteinsviatheirSH2domains.VariousmoleculeswhichinteractwithSHdomainsofNckandregulatesignalingprocessofactinCytoskeletonreorganizationhavebeenidentified.Ncksarethoughttohaveimportantfunctionsinthedevelopmentofmesodermalstructuresduringembryogenesis,linkedtoaroleincellmovementandcytoskeletalreorganization.NckalsohaveafunctioninmodulatingmRNAtranslationatthelevelofinitiationbyinteractingeukayoticinitiationfactor2(eIF2).Underthestressedconditions,proteinsynthesisisreducedbyinhibitingtheactivityofeIF2throughthephosphorylation,transientlyinhibitingrecyclingofeIF2intoitsactiveform.Adapterproteinwhichassociateswithtyrosine-phosphorylatedgrowthfactorreceptorsortheircellularsubstrates.MaintainslowlevelsofEIF2S1phosphorylationbypromotingitsdephosphorylationbyPP1.PlaysaroleintheDNAdamageresponse,notinthedetectionofthedamagebyATM/ATR,butforefficientactivationofdownstreameffectors,suchasthatofCHEK2.
