Mitogen-andstress-activatedproteinkinases(MSK2)isnuclearkinasethatactdownstreamofmitogen-activatedprotein/extracellularsignal-regulatedkinasepathways.ItcontainstwokinasedomainsintheN-terminalandC-terminalregion,respectively.MSK2isactivatedinresponsetomitogenicstimuliviaErk1/2MAPKpathwayandalsobystressstimuliviap38MAPKpathway.Signalsfrommitogensandcellularstressesareinvolvedinmanyfunctionsincludingcellproliferation,differentiation,andsurvivalthroughthephosphorylationofcyclicAMPresponseelement-bindingprotein(CREB)atSer133whichiscatalyzedbyMSK2.Recently,MSK2hasbeenshowntoberequiredforstress-inducedphosphorylationofhistoneH3-Serandtranscriptionalactivationofseveralimmediateearlygenes.Serine/threoninekinasethatmayplayaroleinmediatingthegrowth-factorandstressinducedactivationofthetranscriptionfactorCREB.EssentialroleinthecontrolofRELAtranscriptionalactivityinresponsetoTNF.Phosphorylates"Ser-10"ofhistoneH3inresponsetomitogenics,stressstimuliandepidemalgrowth-factor(EGF)andresultinthetranscriptionalactivationofseveralimmediateearlygenes,includingproto-oncogenesFOSandJUN.Mediatesthemitogen-andstress-inducedphosphorylationofhighmobilitygroupprotein14(HMG-14).
