Mitogen-activatedproteinkinases(MAPKs)areactivatedbyvariousextracellularsignals,suchasgrowthfactors,stresses,andcytokines,andplaycrucialrolesinthedeterminationofcellfatethroughproliferation,differentiation,survival,andapoptosis.Mnk1(MAPkinase-interactingserine/threonine-proteinkinase1)andMnk2areproteinkinasesthataredirectlyphosphorylatedandactivatedbyextracellularsignal-regulatedkinase(ERK)orp38mitogen-activatedprotein(MAP)kinaseswhichphosphorylateatleasttwothreonineresidues(Thr197andThr202ofthemouseMnk1)locatedintheactivationloop.Mnksareimplicatedintheregulationofproteinsynthesisthroughtheirphosphorylationofeukaryotictranslationinitiationfactor4E(eIF4E),alsoknownascap-bindingprotein.Theinitiationstepoftranslationisrate-limitingandrepresentsamajortargetfortranslationalcontrol.Theformationofthecap-bindingcomplexeIF4F,whichisamultiproteinfactorresponsIBLefortherecruitmentofthe40SribosomalsubunittothemRNA5′end,isakeyregulatorystep.Mnk1isphysicallyassociatedwitheIF4FanddirectlyphosphorylateeIF4E,acomponentoftheeIF4Fcomplex.eIF4Especificallybindsthe5′m7GpppNcapstructurefoundinalleukaryoticmRNAsandplaysacriticalroleincap-dependenttranslationinitiationasacentralcomponentoftheeIF4Fcomplex,whichalsocontainseIF4AandeIF4G.Mayplayaroleintheresponsetoenvironmentalstressandcytokines.AppearstoregulatetranscriptionbyphosphorylatingEIF4E,thusincreasingtheaffinityofthisproteinforthe7-methylguanosine-containingmRNAcap.
