TheMAPKinasepathwayisakeysignalingmechanismthatregulatesmanycellularfunctionssuchascellgrowth,proliferation,differentiation,development,transformationandapoptosis.ThebasicarrangementincludesaG-protein,aMAPKkinasekinase(MAPKKK)thatphosphorylatesandactivatesaMAPKkinase(MAPKK),whichinturnphosphorylatesMAPK.TheERK(extracellular-signalregulatedkinase)cascadeisacentralMAPKpathwaywhichcontainsRasasaGprotein,RafasaMAPKKK,MEK1(MAPK/ERKkinase1)andMEK2(MAPK/ERKkinase2)asMAPKK,andERKsasMAPK.MEK1andMEK2are80%identicaltoeachother,andareessentiallyidenticalinmostoftheirkinasedomain.MEK1andMEK2phosphorylateERKsequallywell,bothinvivoandinvitro.MEK1(45KDa)andMEK2(46KDa)arecomposedofacatalytickinasedomain,whichissurroundedbyaregulatoryN-terminaldomain(80aminoacids)andashorterC-terminalregion(30aminoacids).Unlikethekinasedomains,theN-terminiandthecharacteristicPro-richinsertsarequitedivergentbetweenthetwoMEKs(40%identity).MEKsareactivatedbyphosphorylationoftwoSerresiduesintheiractivationloop(Ser218andSer222inMEK1)locatedwithinaSer–Xaa–Ala–Xaa–Ser/Thrmotif,typicaltoallMAPKKs.CatalyzestheconcomitantphosphorylationofathreonineandatyrosineresidueinaThr-Glu-TyrsequencelocatedinMAPkinases.ActivatestheERK1andERK2MAPkinases
