SequentialactivationofproteinkinaseswithintheMAPK(mitogen-activatedproteinkinases)cascadesisacommonmechanismofsignaltransductioninmanycellularprocesses.TheERKsignalingcascadeisacentralMAPKpathwaythatplaysaroleintheregulationofvariouscellularprocessessuchasproliferation,differentiation,development,learning,andsurvival.Mitogen-activatedproteinkinasekinases(MAPKK)phosphorylateMAPK.MEK(MAPkinaseorERKkinase)istheimmediateupstreamactivatorkinaseofERK.ThehumanMEK1encodes393aminoacidresiduesandshares99%aminoacidsequenceidentitywiththemurineMEKland80%withhumanMEK2(400aminoacids).TheMEKsconstituteanevolutionarilyconservedgroupofthreehighlyhomologousmammalianisoforms.Thesearethe45kDaMEK1,itsalternativelysplicedform,MEK1b(43kDa)thatisthoughttobeinactive,andthe46kDaMEK2.Theseproteinsarecomposedofacatalytickinasedomain,whichissurroundedbyaregulatoryN-terminaldomain(80aminoacids)andashorterC-terminalregion(30aminoacids).MEKsareactivatedbyphosphorylationoftwoSerresiduesintheiractivationloop(Ser218andSer222inMEK1).TheactivityofMEKsisregulatedbyadditionalphosphorylation/dephosphorylationprocessesaswell.CatalyzestheconcomitantphosphorylationofathreonineandatyrosineresidueinaThr-Glu-TyrsequencelocatedinMAPkinases.ActivatesERK1andERK2MAPkinases.
