Lipomapreferredpartner(LPP)isencodedbythepreferredfusionpartnergeneofthehighmobilitygroupproteinHMGI-C.LPPisfrequentlyaffectedbychromosomaltranslocationsinamajorgroupofsofttissuelipomas,aswellasinaparosteallipomaandinpulmonarychondroidhamartomas.TheresultingfusionproteinscomprisethreeDNAbindingdomainsofHMGI-CfusedtotwoorthreeC-terminalLIMdomainsofLPPandareciprocalproduct,respectively.ExpressionofaHMGI-C/LPPfusionproteincausesmalignanttransformationoffibroblasts.SimilartoitsrelativeszyxinandTRIP(thyroidreceptorinteractingprotein6)/ZRP-1(zyxin-relatedprotein1),LPPconsistsofaN-terminalproline-richdomainfollowedbythreeC-terminalLIMdomains(doublezincfingerstructuresthatareinvolvedinprotein-proteininteractions).Likethecytoskeletalproteinzyxin,LPPlocalizestofocaladhesionsandcell-celladherensjunctions.LPPharborsanuclearexportsignalanddisplaystranscriptionalactivationcapacity.
