Lactatedehydrogenase(LDH)istheenzymeintheglycolyticpathwaythatconvertspyruvatetolactatewithconcomitantinterconversionofNADHandNAD+.Inmammals,theenzymeisencodedbythreegenes:LDHA(Mormuscleform),LDHB(Horheartform),andLDHC(Xortestisform).ThethreehumanLDHshave84–89%sequencesimilaritiesand69–75%aminoacididentities.TherearefivedifferentLDHisoenzymesbasedontheproportionofMandHchainsexistingintheLDHtetramericstructure.TheLDHAishypoxiainducIBLeanditsexpressionisdirectlycontrolledbythetranscriptionalactivityofthehypoxiainduciblefactor1α(HIF1α).LDHBisalsoknowntobeupregulatedinmanycancers.
