Kininogensareprecursorsforkinin,andthetwomaintypesofthemarehigh-molecularweightkininogen(HMWK)andlow-molecularweightkininogen(LMWK).HMWKalsoknownastheWilliams-Fitzgerald-Flaujeacfactorisaproteinfromthebloodcoagulationsystemaswellasthekinin-kallikreinsystem.Itactsmainlyasacofactoroncoagulationandinflammation,andhasnointrinsiccatalyticactivity.LMWKisproducedlocallybynumeroustissues,andsecretedtogetherwithtissuekallikrein.Kininogensaresynthesizedintheliverandcirculateintheplasmaandotherbodyfluids.Thekininsarepharmacologicallyactivepolypeptidesreleasedinthetissuesandbodyfluidsasaresultoftheenzymaticactionofkallikreinsonkininogens.Thekininfamilyincludesbradykinin(BK)(Arg-Pro-Pro-gly-Phe-Ser-Pro-Phe-Arg),kallidin(Lys-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg)andmethionyl-lysyl-BK(Met-Lys-Arg-Pro-Pro-Gly-Phe-Arg).Kallidinandmethionyl-lysyl-BKareconvertedintoBKbyaminopeptidasespresentinplasmaandurine.ActivetissuekallikreinactsonLMWKtoreleasekallidin.Theplasmakallikreinisfoundincirculationinaninactiveform,whichisknownasprekallikreinorFletcherfactor.BKandkallidinactthroughactivationofG-protein-coupledconstitutiveB(2)orinducIBLekininB(1)receptorslinkedtosignalingpathwaysinvolvingincreasedintracellularCaconcentrationsand/orreleaseofmediatorsincludingarachidonicacidmetabolites,NOandEDHF(Endothelium-derivedhyperpolarizingfactor).Inthecardiovascularsystem,thekallikrein-kininsystemexertsafinecontrolofvascularsmoothmuscletoneandarterialbloodpressure,andplaysasignificantcardioprotectiveeffect.Kininogensareinhibitorsofthiolproteases;(2)HMW-kininogenplaysanimportantroleinbloodcoagulationbyhelpingtopositionoptimallyprekallikreinandfactorXInexttofactorXII;(3)HMW-kininogeninhibitsthethrombin-andplasmin-inducedaggregationofthrombocytes;(4)theactivepeptidebradykininthatisreleasedfromHMW-kininogenshowsavarietyofphysiologicaleffects:(4A)influenceinsmoothmusclecontraction,(4B)inductionofhypotension,(4C)natriuresisanddiuresis,(4D)decreaseinbloodglucoselevel,(4E)itisamediatorofinflammationandcauses(4E1)increaseinvascularpermeability,(4E2)stimulationofnociceptors(4E3)releaseofothermediatorsofinflammation(e.g.prostaglandins),(4F)ithasacardioprotectiveeffect(directlyviabradykininaction,indirectlyviaendothelium-derivedrelaxingfactoraction);(5)LMW-kininogeninhibitstheaggregationofthrombocytes;(6)LMW-kininogenisincontrasttoHMW-kininogennotinvolvedinbloodclotting.
