Thec-JunN-terminalproteinkinases(JNKs)areafamilyofserine/threonineproteinkinasesofthemitogen-activatedproteinkinase(MAPK)group.JNKs,whichareessentialregulatorsofphysiologicalandpathologicalprocesses,areinvolvedinseveraldiseasesincludingdiabetes,atherosclerosis,stroke,andParkinson"sandAlzheimer"sdiseases.TheJNKfamilyconsistsofthreeisoforms;JNK1andJNK2,whichareubiquitous,andJNK3,whichispresentprimarilyintheheart,brainandtestis.Differentialsplicingandexonuseyield10isoformsofJNK.JNK1isanimportantmediatorofinsulinresistanceassociatedwithobesity,butitisalsoindispensablefortheintactcytoarchitectureofthebrain.Respondstoactivationbyenvironmentalstressandpro-inflammatorycytokinesbyphosphorylatinganumberoftranscriptionfactors,primarilycomponentsofAP-1suchasJUN,JDP2andATF2andthusregulatesAP-1transcriptionalactivity.InT-cells,JNK1andJNK2arerequiredforpolarizeddifferentiationofT-helpercellsintoTh1cells.Phosphorylatesheatshockfactorprotein4(HSF4).JNK1isoformsdisplaydifferentbindingpatterns:β-1preferentiallybindstoc-Jun,whereasα-1,α-2,andβ-2haveasimilarlowlevelofbindingtobothc-JunorATF2.However,thereisnocorrelationbetweenbindingandphosphorylation,whichisachievedataboutthesameefficiencybyallisoforms.
