Insulinreceptorsubstrate(IRS)proteinsplayacentralroleinmaintainingbasiccellularfunctionssuchasgrowthandmetabolismthroughinsulin/insulinlikegrowthfactor(IGF)signaling.Fourmembers(IRS-1,IRS-2,IRS-3,IRS-4)ofthisfamilyhavebeenidentifiedwhichdifferintheirsubcellulardistributionandinteractionwithSH2domainproteins.Afterphosphorylationbyactivatedreceptors,theseintracellularsignalingmoleculesrecruitvariousdownstreameffectorpathwaysincludingphosphatidylinositol3-kinase,tyrosineproteinphosphataseSHPTP-2,andseveralsmalleradaptermoleculessuchasthegrowthfactorreceptor-bindingproteinGrb-2.IRS-1,thebestcharacterizedIRSprotein,haseighteenpotentialtyrosinephosphorylationsiteswhichdirectlybindtoSH2domainsindownstremproteins.IRS-1consistsofaminoterminalcontainingpleckstrinhomology(PH)domainfollowedbyaphosphotyrosine-binding(PTB)domainwhichbindstoIRandIGFR,andcarboxyterminalcontainingmultipletyrosineandserineresidueswhichbecomedockingsitesforproteinsthathavePTBdomainsuchasSH2domain.IRS-4isthelastidentifiedmemberoftheIRSfamily.CloningofhumanIRS-4revealedapredictedproteinofsimilarlengthtobothIRS-1andIRS-2andshowedonly27%and29%identitywithIRS-1andIRS-2,respectively.Incontrast,IRS-4exhibitshigherdegreeofhomologyinthePHdomain(43to50%)andthePTBdomain(43to66%)withthecorrespondingdomainsinIRS-1,IRS-2andIRS-3.IRSsarealsothoughttobeabletoinducemalignanttransformation.IRS-1hasbeenshowntobeconstitutivelyactiveinbreastcancer.Actsasaninterfacebetweenmultiplegrowthfactorreceptorspossessingtyrosinekinaseactivity,suchasinsulinreceptor,IGF1RandFGFR1,andacomplexnetworkofintracellularsignalingmoleculescontainingSH2domains.InvolvedintheIGF1Rmitogenicsignalingpathway.PromotestheAKT1signalingpathwayandBADphosphorylationduringinsulinstimulationwithoutactivationofRPS6KB1ortheinhibitionofapoptosis.InteractionwithGRB2enhancesinsulin-stimulatedmitogen-activatedproteinkinaseactivity.Maybeinvolvedinnonreceptortyrosinekinasesignalinginmyoblasts.Playsapivotalroleintheproliferation/differentiationofhepatoblastomacellthroughEPHB2activationuponIGF1stimulation.MayplayaroleinthesignaltransductioninresponsetoinsulinandtoalesserextentinresponsetoIL4andGHonmitogenesis.Playsaroleingrowth,reproductionandglucosehomeostasis.MayactsasnegativeregulatorsoftheIGFIsignalingpathwaybysuppressingthefunctionofIRS1andIRS2.
