IL-36α(IL-1F6),IL-36β(IL-1F8)andIL-36γ(IL-1F9)bindtoIL-36R(IL-1Rrp2)andIL-1RAcP,activatingsimilarintracellularsignalsasIL-1andareinhibitedbyIL-36Ra.TheexpressionofIL-36cytokineshasbeenshowntooccuratdifferentsitesincludingthelungandskinandcanbederivedfromdiversecelltypesincludingkeratinocytes,bronchialepitheliumaswellasmacrophages,monocytesanddifferentTcellsubsets.IL-36familymembersinducetheproductionofproinflammatorycytokines,includingIL-12,IL-1β,IL-6,TNF-αandIL-23inBMDCandCD4Tcells,thuspromotingneutrophilinflux,dendriticcell(DC)activation,polarizationofThelpertype1(Th1)andIL-17-producingTcells(αβTcellsandγδTcells)andkeratinocyteproliferation.Thesecytokinesmayrepresentpotentialtargetsforimmune-mediatedinflammatoryconditionsor,alternatively,couldbeusedasadjuvantsinvaccination.IL-36γisalsoinducedinthelunginvariousmodelsofasthmaandcanbeproducedbybronchialepithelialcellsinresponsetoviralinfection,smokeorinflammatorycytokines.
