Interleukin-33(IL-33;HF-NEV;IL-1F11),amemberoftheIL-1familyofcytokines,isexpressedbymanycelltypesfollowingpro-inflammatorystimulationandisthoughttobereleaseduponcelllysis.IL-33bindstoandsignalsthroughST2(IL-1R1)anditsstimulationrecruitsMYD88,IRAK,IRAK4andTRAF6,followedbyphosphorylationofERK1(MAPK3)/ERK2(MAPK1),p38(MAPK14)andJNK.TheabilityofIL-33totargetnumerousimmunecelltypes,likeTh2-likecells,mastcellsandB1cells,andtoinducecytokineandchemokineproductionunderlinesitspotentialininfluencingtheoutcomeofawiderangeofdiseases,suchasarthritis,asthma,atopicallergy&anaphylaxis,cardiovasculardisease/atherosclerosis,nervoussystemdiseasesandsepsis.
Anti-IL-33,mAb(recombinant)(blocking)(Bondy-1-1)isanantibodydevelopedbyantibodyphagedisplaytechnologyusingahumannaiveantibodygenelibrary.TheselibrariesconsistofscFv(singlechainfragmentvariable)composedofVH(variabledomainofthehumanimmunoglobulinheavychain)andVL(variabledomainofthehumanimmunoglobulinlightchain)connectedbyapolypeptidelinker.Theantibodyfragmentsaredisplayedonthesurfaceoffilamentousbacteriophage(M13).ThisscFvwasselectedbyaffinityselectiononantigeninaprocesstermedpanning.Multipleroundsofpanningareperformedtoenrichforantigen-specificscFv-phage.Monoclonalantibodiesaresubsequentlyidentifiedbyscreeningaftereachroundofselection.TheselectedmonoclonalscFvisclonedintoanappropriatevectorcontainingaFcportionofinterestandthenproducedinmammaliancellstogenerateanIgGlikescFv-Fcfusionprotein.
