IκBkinaseα(IKKα)isacomponentofamultiproteinkinasecomplexthatregulatestheactivityofthetranscriptionfactorNF-κB.ActivationoftheIKKcomplexviaupstreamstimulileadstophosphorylationanddegradationofNF-κBboundIκB(InhibitorofκB).Subsequently,freeNF-κBdimersenterthenucleusandregulatethetranscriptionofavarietyoftargetgenesinvolvedincellproliferationanddifferentiation,apoptosis,andinflammation,etc.TheIKKcomplexconsistsof2highlyhomologouskinasesubunits,IKKαandIKKβ,andanonenzymaticregulatorycomponent,IKKγ/NEMO.TherelativecontributionsofIKKαandIKKβtothesignallingcomplexvaryaccordingtotherequirementsofthecell.IKKβplaysapredominantroleinimmuneresponses,whileIKKαaloneappearstobesufficientforatleastsomedevelopmentalsystems.Recently,IKK/NF-κBsignalingpathwaywasstudiedastherapeutictargetsincancer.ActsaspartoftheIKKcomplexintheconventionalpathwayofNF-κ-BactivationandphosphorylatesinhibitorsofNF-κ-BthusleADIngtothedissociationoftheinhibitor/NF-κ-Bcomplexandultimatelythedegradationoftheinhibitor.Aspartofthenon-canonicalpathwayofNF-κ-Bactivation,theMAP3K14-activatedCHUK/IKKAhomodimerphosphorylatesNFKB2/p100associatedwithRelB,inducingitsproteolyticprocessingtoNFKB2/p52andtheformationofNF-κ-BRelB-p52complexes.AlsophosphorylatesNCOA3.Phosphorylates"Ser-10"ofhistoneH3atNF-κ-B-regulatedpromotersduringinflammatoryresponsestriggeredbycytokines.
