IA-2(insulinoma-associatedprotein2,ICA512,PTPRN)isamemberofthetransmembraneproteintyrosinephosphatase(PTP)familylocatedinsecretorygranulesofneuroendocrinecells.TheIA-2proteinis979aminoacidsinlengthandconsistsofaluminaldomain,transmembranedomain,andcytoplasmicdomain.AlthoughamemberofthePTPfamily,IA-2lacksphosphataseactivitywithknownsubstratesduetoaminoacidsubstitutionsatcriticalsitesinitsPTPdomain.TwoparalogRPTPs,IA-2andIA-2βwereidentifiedasmajorautoantigensintype-1diabetesmellitus.Ongranuleexocytosis,theIA-2cytoplasmicdomainiscleavedandtheresultingcytosolicfragmentmovesintothenucleuswhereitenhancesthelevelsofphosphorylatedSTAT5andSTAT3,therebyinducinginsulingenetranscriptionandgranulebiogenesis.IA-2signalingenhancespancreaticβ–cellproliferationbyregulatingcyclinsDthroughSTATs.Implicatedinneuroendocrinesecretoryprocesses.Maybeinvolvedinprocessesspecificforneurosecretorygranules,suchastheirbiogenesis,traffickingorregulatedexocytosisormayhaveageneralroleinneuroendocrinefunctions.Seemstolackintrinsicenzymeactivity.MayplayaroleintheregulationofsecretorygranulesviaitsinteractionwithSNTB2.
