Heatshockproteinsareubiquitousproteinsandhavebeencharacterizedascytoprotectivemolecularchaperones.Thetypicalfunctionofachaperoneistoassistaproteintoattainitsfunctionalconformationtopreventnon-functionalaggregationofmisfoldedproteins.TheprincipalHSPfamiliesareHSP90,HSP70,HSP60andthesmallHSPsincludingHSP27,ubiquitin,α-crystallin,Hsp20andothers.ThecommonfunctionsofsmallHspsarechaperoneactivity,Thermotolerance,inhibitionofapoptosis,regulationofcelldevelopment,andcelldifferentiation.Hsp27hasamolecularweightofapproximately27kDa,althoughithasbeenshowntoformlargeaggregatesofupto800kDainthecytosol.Hsp27isfoundinseveraltypesofhumancells,includingtumourcells.Hsp27interfereswithapoptosisthroughitsabilitytointeractwithandinhibitkeycomponentsoftheapoptoticsignalingpathway,includingthecaspaseactivationcomplex.OverexpressionofheatshockproteinscanincreasethetumOrigenicpotentialoftumourcells.HSP27alsohasbeenreportedtobeinvolvedindevelopmentandprogressionofhormone-refractoryprostatecancer.Involvedinstressresistanceandactinorganization.
