HMGB1wasoriginallydiscoveredasanessentialDNA-bindingproteinforregulatingp53,NF-κBandotherimportantproteins.Itissecretedfromactivateddentriccells,macrophageandnectroticcells,andactsasaligandforRAGE,TLR-2andTLR-4expressedonsurroundingcells.Asaresult,HMGB1activatesRac,CDC42andNF-κBinducinglocalizedinnateimmunityofdamagedtissue,tissueregenerationbyrecruitmentofstemcellsandhemostasisbyinductionoftissuefactorexpression.HMGB1isalsoacausativeagentofvariousdiseasesasitcauseslocalizedinflammationsuchasarteriosclerosis,chronicrheumatoidarthritisandnephritis.
Anti-HMGB1,mAb(recombinant)(Giby-1-4)(Biotin)isanantibodydevelopedbyantibodyphagedisplaytechnologyusingahumannaiveantibodygenelibrary.TheselibrariesconsistofscFv(singlechainfragmentvariable)composedofVH(variabledomainofthehumanimmunoglobulinheavychain)andVL(variabledomainofthehumanimmunoglobulinlightchain)connectedbyapolypeptidelinker.Theantibodyfragmentsaredisplayedonthesurfaceoffilamentousbacteriophage(M13).ThisscFvwasselectedbyaffinityselectiononantigeninaprocesstermedpanning.Multipleroundsofpanningareperformedtoenrichforantigen-specificscFv-phage.Monoclonalantibodiesaresubsequentlyidentifiedbyscreeningaftereachroundofselection.TheselectedmonoclonalscFvisclonedintoanappropriatevectorcontainingaFcportionofinterestandthenproducedinmammaliancellstogenerateanIgGlikescFv-Fcfusionprotein.
