Hemoglobin(haemoglobin,Hb)istheiron-containingoxygen-transportproteinintheredbloodcellsofvertebrates.Itschiefphysiologicalfunctionistotransportoxygenfromthelungsorgillstothetissueswhereitreleasestheoxygenforthecellularrespiration.HbA(mostofadulthemoglobin)isatetramericproteinwithamolecularweightof64,500Da,consistingoftwoidenticalα-chainsof141aminoacidseach,andtwoidenticalβ-chainsof146aminoacidseach.EachsubunitorchainofHbAadoptsathreedimensionalstructure,calledtheglobinfold,similartothatoftherelated,monomericproteinmyoglobin(Mb).Thisfoldisanarrangementoftheheliceswhichformsapocketthatenclosesandbindsahemeprostheticgroupwhichconsistsofaniron(Fe)ionheldinaheterocyclicring,knownasaporphyrin.Thebindingofoxygenisacooperativeprocess.Thebindingaffinityofhemoglobinforoxygenisincreasedbytheoxygensaturationofthemolecule.theoxygenbindingcurveofhemoglobinissigmoidal,orS-shaped,asopposedtothenormalhyperboliccurveassociatedwithnoncooperativebinding.Hemoglobin"soxygen-bindingcapacityisdecreasedinthepresenceofcarbonmonoxidebecausebothgasescompeteforthesamebindingsitesonhemoglobin.Carbondioxideoccupiesadifferentbindingsiteonthehemoglobin.Somemutationsintheglobinchainareassociatedwiththehemoglobinopathies,suchassickle-celldiseaseandthalassemia.Hemoglobin(Hb)-basedO2carriers(HBOCs)suchashemoglobinvesicle(HbV)havebeeninvestigatedasatransfusionalternativeandotherclinicalapplications.Involvedinoxygentransportfromthelungtothevariousperipheraltissues.LVV-hemorphin-7potentiatestheactivityofbradykinin,causingadecreaseinbloodpressure.
