GPI-PLD(glycosylphosphati-dylinositol-specificphospholipaseD),a815-aminoacidprotein,isexpressedinnumeroustissuesandcellsandspecificallycleavesGPI-anchoredproteins.LiverhasthehighestlevelofGPI-PLDexpressionandistheprimaryorgancontributingtoGPI-PLDintheserum.GPI-PLDisabundantinseruminwhichitassociateswithapolipoproteinsAIandAIV.IncreasedserumGPI-PLDisassociatedwithinsulinresistanceandelevatedserumtriglycerides.Manysurfaceproteinsareattachedtoeukaryoticcellmembranesviaglycosylphosphatidylinositol(GPI)anchorsthatarecovalentlyboundtotheC-terminusoftheproteinandcleavageoftheGPImoietybyGPI-PLD,onlyenzymeknownthatcleavageGPIanchor,mayrepresentameansofregulatingattachmentoftheseproteinstothecellsurface,oralternatively,theirreleaseintotheextracellularenvironment.Thisproteinhydrolyzestheinositolphosphatelinkageinproteinsanchoredbyphosphatidylinositolglycans(GPI-anchor)thusreleasingtheseproteinsfromthemembrane.
