Glutathionereductase(GR)isamemberofpyridinenucleotide-disulfideoxidoreductases,whichincludesthecloselyrelatedenzymesthioredoxinreductase,lipoamidedehydrogenase,trypanothionereductaseandmercuricionreductase.GRisacytoplasmicflavoenzymewidelydistributedinaerobicorganisms.Thedimericproteiniscomposedoftwoidenticalsubunits,eachcontaining1FADand1redox-activedisulfide/dithiolascomponentsofthecatalyticapparatus.Itplaysaroleinmaintainingglutathione(GSH)initsreducedformbycatalyzingthereductionofglutathionedisulfide(GSSG)(1):GSSG+NADPH+H+⇔2GSH+NADP+Inmosteukaryoticcells,GRmaintainstheratioof[GSH]/[GSSG]elevated,andparticipatesinseveralvitalfunctionssuchasthedetoxificationofreactiveoxygenspeciesaswellasproteinandDNAbiosynthesis(2).Maintainshighlevelsofreducedglutathioneinthecytosol.
