Glutathioneperoxidases(Gpxs)areubiquitouslyexpressedproteinswhichcatalyzethereductionofhydrogenperoxidesandorganichydroperoxidesbyglutathione.Thereareseveralisoformswhichdifferintheirprimarystructureandlocalization.Theclassicalcytosolic/mitochondrialGPx1(cGPx)isaselenium-dependentenzyme,firstoftheGPxfamilytobediscovered.GPx2,alsoknownasgastrointestinalGPx(GI-GPx),isanintracellularenzymeexpressedonlyattheepitheliumofthegastrointestinaltract(1).ExtracellularplasmaGPx(pGPxorGPx3)ismainlyexpressedbythekidneyfromwhereitisreleasedintothebloodcirculation(2).PhospholipidhydroperoxideGPx4(PH-GPx)expressedinmosttissues,canreducemanyhydroperoxidesincludinghydroperoxidesintegratedinmembranes,hydroperoxylipidsinlowdensitylipoproteinorthymine(3).AllmammalianGPxfamilymembers,exceptfortherecentlydescribedCyscontainingGPx3andepididymis-specificsecretoryGPx(eGPxorGPx5)isoforms,possessselenocysteineattheactivesite(4-5).
