Glutaredoxin(Grx),alsoknownasthioltransferase,isasmallheat-stableoxidoreductase.Grxsformpartoftheglutaredoxinsystem,comprisingNADPH,GSHandglutathionereductase,whichtransferselectronsfromNADPHtoglutaredoxinsviaGSH(1).FirstrecoveredinE.coliasGSH-dependenthydrogendonorsforribonucleotidereductase,GrxcatalyzesGSH-disulfideoxido-reductaseviatworedox-activecysteineresidues(2).Theactivesequence(Cys-Pro-Tyr-Cys)isconservedinavarietyofspecies.The12kDadithiolproteinhasaroleinreductionofmixeddisulfidesincellsexposedtooxidativestress(3).Hasaglutathione-disulfideoxidoreductaseactivityinthepresenceofNADPHandglutathionereductase.Reduceslowmolecularweightdisulfidesandproteins.
