GlutamineSynthetase(GS)catalyzestheconversionofammoniaandglutamatetoglutamine.ThisreactionconsumesamoleculeofATP:Glutamate+NH4++ATP>>Glutamine+ADP+Pi.GSisfoundinastrocytesasanoctamerofidentical45kDasubunits.MostwellknownfunctionofGSisthedetoxificationofbrainammonia.Italsohasanimportantroleincontrollingmetabolicregulationsofneurotransmitterglutamate.BecauseofthemultiplefunctionsandimportanceofGSincellularmetabolism,bothcatalyticactivitiesandsynthesisarehighlyregulated.TheactivityofGSiscontrolledbyadenylylation.ItsactivityisdecreasedinthecerebralcortexofbrainsaffectedbyAlzheimer’sdisease,particularlyinthevicinityofsenileplaques.Itisalsodecreasedunderconditionsofglucosedeprivation.Ontheotherhands,thelevelofexpressionofGSisincreasedduringischemiainvivoorhypoxiainculture.Essentialforproliferationoffetalskinfibroblasts.
