Gelsolinsuperfamilyproteinscontrolactinorganizationbyseveringfilaments,cappingfilamentendsandnucleatingactinassembly.Gelsolin,aproteinof82–84kDa,isthefoundingmemberofthisfamily,whichnowcontainsatleastanothersixmembers:villin,adseverin,capG,advillin,supervillinandflightlessI.Gelsolinexistsasacytoplasmicaswellasaplasmaisoform,andcanbind,severandcapactinfilaments.Plasmagelsolinleveldecreasesdramaticallyasaresultofmajortraumaandreinfusionofgelsolincanprotectagainstlungdamageassociatedwithmajorburninjuryandothertypesofinsults.RecentstudyinFasantibody-inducedliverfailuresuggeststhatgelsolinexertsanoverallanti-apoptoticeffectinvivo.Gelsolin,aMarkerofmotility,couldbeappliedasabiomarkerinassessingtumorprognosis.Tumor-associatedprocessessuchasinvasionandmetastasisareknowntobecriticallydependentondynamicalterationsintheorganizationoftheactinCytoskeleton.AcausalrelationshipbetweengelsolinexpressionandinvitroinvasionbywayofsignalingthroughRashasbeenreported.Calcium-regulated,actin-modulatingproteinthatbindstotheplus(orbarbed)endsofactinmonomersorfilaments,preventingmonomerexchange(end-blockingorcapping).Itcanpromotetheassemblyofmonomersintofilaments(nucleation)aswellasseverfilamentsalreadyformed.
