Glyceraldehyde-3-phosphatedehydrogenase(GAPDH)isacatalyticenzymecommonlyknowntobeinvolvedinglycolysis.Theenzymeexistsasatetramerofidentical37-kDasubunits.GAPDHcatalyzesthereversIBLereductionof1,3-bisphosphoglyceratetoglyceraldehyde3-phosphophateinthepresenceofNADPH.Apartfromplayingakeyroleinglycolysis,thisubiquitouslyexpressedenzymealsodisplaysotheractivitiesunrelatedtoitsglycolyticfunction.GAPDHisreportedtobeinvolvedintheprocessesofDNAreplication,DNArepair,nuclearRNAexport,membranefusionandmicrotubulebundling.OtherstudiesalsoprovideevidenceofGAPDHplayinganessentialpartoftheprogramofgeneexpressionobservedinapoptosisandaspartofthecellularphenotypeofage-relatedneurodegenerativediseases.Onrecentstudy,GAPDHhasidentifiedofthemostoxidantsensitivecellproteins.Independentofitsglycolyticactivityitisalsoinvolvedinmembranetraffickingintheearlysecretorypathway.
