Glyceraldehyde-3-phosphatedehydrogenase(GAPDH)isacatalyticenzymecommonlyknowntobeinvolvedinglycolysis.Theenzymeexistsasatetramerofidentical37kDasubunits.GAPDHcatalyzesthereversIBLereductionof1,3-bisphosphoglyceratetoglyceraldehyde3-phosphophateinthepresenceofNADPH.Apartfromplayingakeyroleinglycolysis,thisubiquitouslyexpressedenzymealsodisplaysotheractivitiesunrelatedtoitsglycolyticfunction.GAPDHisreportedtobeinvolvedintheprocessesofDNAreplication(1),DNArepair(2),nuclearRNAexport(3-4),membranefusion(5)andmicrotubulebundling.OtherstudiesalsoprovideevidenceofGAPDHplayinganessentialpartoftheprogramofgeneexpressionobservedinapoptosisandaspartofthecellularphenotypeofage-relatedneurodegenerativediseases(6-7).Hasbothglyceraldehyde-3-phosphatedehydrogenaseandnitrosylaseactivities,therebyplayingaroleinglycolysisandnuclearfunctions,respectively.Participatesinnucleareventsincludingtranscription,RNAtransport,DNAreplicationandapoptosis.NuclearfunctionsareprobablyduetothenitrosylaseactivitythatmediatescysteineS-nitrosylationofnucleartargetproteinssuchasSIRT1,HDAC2andPRKDC.Glyceraldehyde-3-phosphatedehydrogenaseisakeyenzymeinglycolysisthatcatalyzesthefirststepofthepathwaybyconvertingD-glyceraldehyde3-phosphate(G3P)into3-phospho-D-glyceroylphosphate.
