Theepidermalgrowthfactorreceptor(EGFR)isatransmembranereceptortyrosinekinaseoftheErbB(alsoknownasHER)familyinwhichfourmembershavebeenidentified:EGFR(ErbB1),HER2/Neu(ErbB2),HER3(ErbB3),andHER4(ErbB4).AllfourerbBreceptorsarecomposedofanextracellularligand-bindingregionconsistingofglycosylateddomains,atransmembranedomaincontainingasinglehydrophobicanchorsequence,anintracellularregioncontainingthecatalytictyrosinekinasedomain,andacarboxyl-terminalregioncontainingseveraltyrosineresiduesthatbecomephosphorylatedafterreceptoractivation.Theepidermalgrowthfactorreceptor(EGFR)signalingpathwayisoneofthemostimportantpathwaysthatregulategrowth,survival,proliferation,anddifferentiationinmammaliancells.EGFRandothermembersoftheerbBfamilyformeitherhomodimersorheterodimersuponligandbinding,resultinginconformationalchangesthatallowactivationofproteinkinasesandtransphosphorylationofkeytyrosineresidueswithinthecarboxyl-terminaldomain.Aftertheinductionoftyrosinephosphorylation,somesignalingpathwaysappeartostartwiththerecognitionoftheC-terminalphosphotyrosinesbyappropriateadaptororsignalingmolecules.TheaberrantactivationoftheEGFRleadstoenhancedproliferationandothertumor-promotingactivities.Severalmechanismsleadtoaberrantreceptoractivation,includingreceptoroverexpression,geneamplification,activatingmutations,overexpressionofreceptorligands,and/orlossoftheirnegativeregulatorymechanisms.Theepidermalgrowthfactorreceptor(EGFR)hasbeenextensivelyinvestigatedasatargetforanti-neoplastictherapy.Anti-EGFRantibodiesthatinterferewithligand-dependentreceptoractivationhaveshownclinicalactivityinavarietyofsolidtumors.ReceptorforEGF,butalsoforothermembersoftheEGFfamily,asTGF-α,amphiregulin,βcellulin,heparin-bindingEGF-likegrowthfactor,GP30andvacciniavirusgrowthfactor.Isinvolvedinthecontrolofcellgrowthanddifferentiation.PhosphorylatesMUC1inbreastcancercellsandincreasestheinteractionofMUC1withSRCandCTNNB1/β-catenin.Isoform2mayactasanantagoNISTofEGFaction.
