DiazepamBindingInhibitor(DBI)isahighlyconserved10kDapolypeptidewhichisexpressedinvariousspeciesrangefromyeasttomammals.AsaninverseagoNISTforbenzodiazepinereceptors,DBIdownregulatesinhibitoryeffectsofGABA.Italsohaspotentialtoinduceanxiety.Foundincentralandperipheraltissues,DBIalsoparticipatesinmetabolismofsteroids,whichhasbeenknowntopartiallymodifyGABAAreceptorfunctionintheCNS.Inperipheraltissues,DBIplaysregulatoryrolesinsteroidogenesis.DBIlevelshavebeenreportedtobedecreasedinthecerebrospinalfluidobtainedfrompatientswithAlzheimerdisease.Bindsmedium-andlong-chainacyl-CoAesterswithveryhighaffinityandmayfunctionasanintracellularcarrierofacyl-CoAesters.Itisalsoabletodisplacediazepamfromthebenzodiazepine(BZD)recognitionsitelocatedontheGABAtypeAreceptor.ItisthereforepossIBLethatthisproteinalsoactsasaneuropeptidetomodulatetheactionoftheGABAreceptor.
