CylindromasarebenignadnexalskintumorscausedbygermlinemutationsintheCYLDgene.InmostcasesthesecondwildtypealleleislostintumortissuesuggestingthatCYLDfunctionsastumor-suppressor.CYLDisaproteinof956aminoacidsharboringafunctionaldeubiquitinatingdomainattheC-terminalend.CYLDwasshowntointeractwithTRAF2,NEMOandTRIP.FurThermore,CYLDisanegativeregulatorofNF-κBactivationmediatedbyTNFfamilymembersandIL-1β.DownregulationofNF-κBactivitydependsonthedeubiquitinatingactivityofCYLDwhichremoveslysine-63linkedubiquitinchainsfromtargetproteins.ThesefindingsindicatethatcylindromasarisethroughconstitutiveNF-κBactivationleADIngtohyperproliferationandtumorgrowth.Insummary,CYLDcontributestotheregulationofcellsurvival,proliferationanddifferentiationviaitseffectsonNF-κBactivation.ItisanegativeregulatoroftheWntsignaling,inhibitsHDAC6andtherebypromotesacetylationofα-tubulinandstabilizationofmicrotubules.Throughtheregulationofmicrotubuledynamics,itcontributestotheregulationofcellproliferation,cellpolarization,cellmigrationandangiogenesis.ItplaysaroleintheregulationofinflammationandtheinnateimmuneresponseviaitseffectsonNF-κBactivation.
