Cellularretinaldehyde-bindingprotein(CRALBP)isacytoplasmicprotein,abundantlyexpressedintheretinalpigmentepithelium(RPE)andMüllergliaoftheretinaandinthepinealgland.Structurally,humanCRALBPisa∼36-kDamonomericprotein,proposedtoadoptan“open”or“closed”conformation,dependingonwhetheritiscarryinganendogenousligand.CRALBPinteractsstructurallyandfunctionallywith11-cis-retinoldehydrogenase(RDH5),anenzymeofthevisualcycleinRPE.CRALBPisamemberoftheCRAL_TRIOfamilyofproteinsthatsharealipid-bindingdomainderivedfromtheyeastSec14protein.MutationsinthehumanCRALBPgenecauseretinalpathologyanddelayeddarkadaptation.CRALBPknockoutmicehaveadelayedresponseinrhodopsinregeneration,11-cis-retinalproductionanddarkadaptationafterIllumination.Solubleretinoidcarrieressentialtheproperfunctionofbothrodandconephotoreceptors.Participatesintheregenerationofactive11-cis-retinoland11-cis-retinaldehyde,fromtheinactive11-transproductsoftherhodopsinphotocycleandinthedenovosynthesisoftheseretinoidsfrom11-transmetabolicprecursors.Thecyclingofretinoidsbetweenphotoreceptorandadjacentpigmentepitheliumcellsisknownasthe"visualcycle".
