Thecomplementsystemisapartofthelargerimmunesystemandthreebiochemicalpathwaysarepresent:theclassicalcomplementpathway,thealternativepathway,andthemannose-bindinglectinpathway.HumancomplementfactorC6isoneoffivecomponents(C5b,C6,C7,C8,andC9)thatinteracttoformthecytolyticmembraneattackcomplex(MAC)whichisthecytolyticendproductofthecomplementcascade.MACistypicallyformedonthesurfaceofintrudingpathogenicbacterialasaresultoftheactivationofthecomplementsystem,anditisoneoftheultimateweaponsoftheimmunesystem.Thesixthcomponentofcomplement,C6,isa913aminoacidsinglepolypeptidechainserumglycoprotein.HomologystudysuggeststhatC6couldcontaintwodomains,anamino-terminalregionthatisrelatedtocomplementC8andC9,andacarboxyl-terminalregionthathaspartialhomologytothecomplementregulatoryproteinsfactorHandfactorI.GeneticdeficienciesofterminalcomplementcomponentsleadtomarkedlyincreasedsusceptibilitytoonlyoneparticularGram-negativegenus,theNeisseria.Thesusceptibilityisattributabletothemajorroleplayedbycomplement-mediatedkillinginhostdefenseagainstthepathogen.Constituentofthemembraneattackcomplex(MAC)thatplaysakeyroleintheinnateandadaptiveimmuneresponsebyformingporesintheplasmamembraneoftargetcells.
