Thecomplementsystemisapartofthelargerimmunesystemandthreebiochemicalpathwaysarepresent:theclassicalcomplementpathway,thealternativepathway,andthemannose-bindinglectinpathway.ComplementcomponentC4isanessentialcomponentofhumoralimmuneresponse.Initsactivatedform,C4bbecomesasubunitoftheC3convertase,whichisanenzymaticcomplexthatactivatesC3oftheclassicalandlectincomplementactivationpathways.TheclassicalpathwayisinitiatedbytheactivationoftheC1-complex(C1q,C1randC1s)byC1q"sbindingtoantibody-antigen.TheC1-complexnowbindstoandsplitsC2andC4producingC2aandC4b.C4bandC2abindtoformC3-convertase.ProductionofC3-convertaseleadstocleavageofC3intoC3aandC3bandC3bjoinswiththeC3convertasetomakeC5convertase.HumanC4isthemostpolymorphicproteinofthecomplementsystem.ComplementC4existsastwoisotypes,C4A(acidic)andC4B(basic).Althoughthesequenceidentityisveryhigh,theyhavedifferenthemolyticactivities,covalentaffinitiestoantigensandimmunecomplexes,andSEROlogicalreactivities.EachC4containsβchain,αchain,C4aanaphyltoxin,C4b,andγchain.C4-deficientmiceshowsincompleteclearanceofmicrobialattackandC4-deficiencyinhumanshowsincreasedautoimmunediseases.C4playsacentralroleintheactivationoftheclassicalpathwayofthecomplementsystem.ItisprocessedbyactivatedC1whichremovesfromtheαchaintheC4aanaphylatoxin.TheremainingαchainfragmentC4bisthemajoractivationproductandisanessentialsubunitoftheC3convertase(C4b2a)andtheC5convertase(C3bC4b2a)enzymesoftheclassicalcomplementpathway.DerivedfromproteolyticdegradationofcomplementC4,C4aanaphylatoxinisamediatoroflocalinflammatoryprocess.Itinducesthecontractionofsmoothmuscle,increasesvascularpermeabilityandcauseshistaminereleasefrommastcellsandbasophilicleukocytes.
