Theproteolysisofcollagensplaysanimportantroleinnumerousphysiologicalandpathologicalsituationssuchasmorphogenesis,woundhealing,arthritis,arteriosclerosisandtumormetastasis.TriplehelicaltypeIcollagensaremadeupoftwoα1(I)andoneα2(I)chains,andarefoundinskin,tendon,ligamentandinterstitialtissues.Duetotheirfibrillarystructurenativecollagensareresistanttomostproteases.Theyaresubstratesforcertainmatrixmetalloproteinases(MMPs),whichconstituteafamilyofzinc-dependentenzymescatalyzingthedegradationofextracellularmatrixcomponents.InitialMMP-8dependentcleavageofcollagenintothecharacteristic¾and¼fragmentshasbeenshowntoenableMMP-9diffusionalongtheproteinhelixwithpreferentialbindingtothecollagen¾fragmenttail.Finally,untwistingofthehelixendresultsinthelocaldenaturationofthetriplehelicalstructure.
