BothChk1andChk2arecriticallyimportantcheckpointkinases.Chk1isessentialfornormaldevelopmentandcellgrowthandChk1deficiencyinmouseorinembryonicstem(ES)cellsislethal.LossofChk2istoleratedandhasbeenfoundtobeassociatedwithasubsetofLi-Fraumenicases.Li-Fraumenisyndromeincreasesgreatlythesusceptibilitytocancer,withparticularlyhighoccurrencesofbreastcancer,braintumors,acuteleukemia,softtissuesarcomas,bonesarcomas,andadrenalcorticalcarcinoma.Chk2regulatescellcyclecheckpointsandapoptosisinresponsetoDNAdamage,particularlytoDNAdouble-strandbreaks.Thecelleitherstopsthecellcycleandthereforeitsproliferationuntilthedamageisrepaired,oritdestructsitself(apoptosis).Theataxiatelangiectasiamutated(ATM)andATR(ATMandRad3-related)proteinkinasesexertcellcycledelay,inpart,byphosphorylatingChk1,Chk2,andp53.PhosphorylationonChk1andp53requiresATR,whereasphosphorylationonChk2requiresATM.ScreeningofnovelsubstratesofChk1andCkh2followedbyimunoprecipitationkinaseassayandsite-directedmutagenesisanalysissuggeststhatHDAC5andPGC-1arespecifictargetsforChk1and/orChk2atleastinvitro.RegulatescellcyclecheckpointsandapoptosisinresponsetoDNAdamage,particularlytoDNAdouble-strandbreaks.InhibitsCDC25Cphosphatasebyphosphorylationon"Ser-216",preventingtheentryintomitosis.Mayalsoplayaroleinmeiosis.RegulatestheTP53tumorsuppressorthroughphosphorylationat"Thr-18"and"Ser-20".
