Procaspase-8belongstothefamilyofcaspases.BindingofFasLtoFasleadstoformationofareceptorcomplexatthecellularmembrane,whichwasnamedDISC.TheDISCconsistsofoligomerizedreceptors,theDD-containingadaptormoleculeFADD,procaspase-8,procaspase-10andc-FLIP.TheDISCstructureprovidesaplatformfortheoligomerizationofprocaspase-8thatallowstwoprocaspase-8homodimerstobeinthecloseproximityleADIngtotheinitialactivationofprocaspase-8.Atthefirstcleavagestep,theN-terminalp43/p41andtheC-terminalp30cleavageproductsaregenerated.Importantly,thesecleavageproductsalreadypossesscatalyticactivity.Atthesecondcleavagestep,p43/p41andp30areprocessedtop10andp18,respectively,whichleadstothegenerationoftheactivecaspase-8heterotetramer(p18/p10)2.
