Caspase-activatedDNase(CAD)cleaveschromosomalDNAduringapoptosis.Cancerwhichisacelldeathprocessremovingtoxicand/oruselesscellsduringmammaliandevelopmentcanbeinducedbyavarietyofstimuli,includingthedeathfactorsofthetumornecrosisfactorfamily,orUVirrADIation,anticancerdrugs,andsurvivalfactordeprivation.TheapoptoticprocessisaccompaniedbyshrinkageandfragmentationofthecellsandnucleianddegradationofthechromosomalDNAintonucleosomalunits.CAD,whichcanbeactivatedbycaspases,existsasacomplexwithitsinhibitor,inhibitorofCAD(ICAD).ICADhelpscorrectfoldingofCADandremainscomplexedwithCADtoinhibittheDNaseactivityofCAD.Caspase,whichisactivatedduringtheapoptoticprocess,cleavesICAD,andtheCADreleasedfromICADentersthenucleitocleavethechromosomalDNA.Internalaswellasexternalsignalscanactivatecaspasesandthisisunderthecontrolofthebalancebetweenpro-andanti-apoptoticproteinsoftheBcl-2family,heatshockproteins,andinhibitorsofapoptosisproteins(IAPs).Currently,fourteenmembersofthecaspasefamilyhavebeenidentified,ofwhichsevenmediateapoptosis.NucleasethatinducesDNAfragmentationandchromatincondensationduringapoptosis.DegradesnakedDNAandinducesapoptoticmorphology.
