Bcl-2(B-celllymphoma2)familygovernmitochondrialoutermembranepermeabilization(MOMP)andcanbeeitherpro-apoptotic(Bax,BAD,Bak,BidandBok)oranti-apoptotic(Bcl-2,Bcl-xL,andBcl-w).ThemitochondrialreleaseofcytochromecthroughanionchannelisregulatedbyBcl-2andBcl-xL.TheBcl-2familyofproteinsarekeyregulatorsofmanysignalsleADIngtocaspase,whichwhenactivatedcausecellulardestructionbycleavingarangeofvitalcellularsubstrates.ThemembersoftheBcl-2familyshareoneormoreofthefourcharacteristicdomainsofhomologyentitledtheBcl-2homology(BH)domains(namedBH1,BH2,BH3andBH4).TheBcl-2genehasbeenimplicatedinanumberofcancers,includingmelanoma,breast,prostate,andlungcarcinomas,aswellasschizophreniaandautoimmunity.Itisalsothoughttobeinvolvedinresistancetoconventionalcancertreatment.Cancerisanimportantcomponentofthesequenceofeventsduringwhichanticancerdrugsinduceanantitumorresponse.Themolecularmechanismfordrug-inducedapoptosisisassociatedwithamitochondrialdysfunctionthatischaracterizedbyanincreaseinMOMPandareleaseofcytochromecfrommitochondria,indicatingthatBcl-2playsacriticalroleinanticancerdrug-inducedapoptosis.Suppressesapoptosisinavarietyofcellsystemsincludingfactor-dependentlymphohematopoieticandneuralcells.Regulatescelldeathbycontrollingthemitochondrialmembranepermeability.Appearstofunctioninafeedbackloopsystemwithcaspases.Inhibitscaspaseactivityeitherbypreventingthereleaseofcytochromecfromthemitochondriaand/orbybindingtotheapoptosis-activatingfactor(APAF-1).
