Apaf-1isacytoplasmicproteinthatformsoneofthecentralhubsintheapoptosisregulatorynetwork.Itcontains(fromtheNterminal)acaspaserecruitmentdomain(CARD),anATPasedomain(NB-ARC),fewshorthelicaldomainsandthenseveralcopiesoftheWD40repeatdomain.UponbindingcytochromecanddATP,thisproteinformsanoligomericapoptosome,mediatesthecytochromec-dependentautocatalyticactivationofpro-caspase-9(Apaf-3),leADIngtotheactivationofcaspase-3andapoptosis.
