Angiopoietin-1(Ang-1)andAngiopoietin-2(Ang-2)arecloselyrelatedsecretedligandswhichbindwithsimilaraffinitytoTie-2.Tie-2andangiopoietinshavebeenshowntoplaycriticalrolesinembryogenicangiogenesisandinmaintainingtheintegrityoftheadultvasculature.Ang-1activatesTie-2signalingonendothelialcellstopromotechemotaxis,cellsurvival,cellsprouting,vesselgrowthandstabilization.Ang-2hasbeenidentifiedasasecretedproteinligandofTie-2andhasalternativelybeenreportedtobeanantagoNISTforAng-1inducedTie-2signalingaswellasanagonistforTie-2signaling,dependingonthecellcontext.
anti-Angiopoietin-2,mAb(rec.)(blocking)(Angy-2-1)(preservativefree)isanantibodydevelopedbyantibodyphagedisplaytechnologyusingahumannaiveantibodygenelibrary.TheselibrariesconsistofscFv(singlechainfragmentvariable)composedofVH(variabledomainofthehumanimmunoglobulinheavychain)andVL(variabledomainofthehumanimmunoglobulinlightchain)connectedbyapolypeptidelinker.Theantibodyfragmentsaredisplayedonthesurfaceoffilamentousbacteriophage(M13).ThisscFvwasselectedbyaffinityselectiononantigeninaprocesstermedpanning.Multipleroundsofpanningareperformedtoenrichforantigen-specificscFv-phage.Monoclonalantibodiesaresubsequentlyidentifiedbyscreeningaftereachroundofselection.TheselectedmonoclonalscFvisclonedintoanappropriatevectorcontainingaFcportionofinterestandthenproducedinmammaliancellstogenerateanIgGlikescFv-Fcfusionprotein.
