α2-Macroglobulin(α2M),isa720-kDahomotetramericglycoproteincomposedoffouridentical180kDasubunit.α2Mshareswithotherα-macroglobulins,likethecomplementcomponentsC3andC4andthepregnancyzoneproteinPZP,anextraordinarybindingcapacityforavarietyofligands.Thisallowstheα-macroglobulinstoserveashumoraldefensebarriersagainstforeignpeptidesintheplasma.α2Minteractsandcapturesvirtuallyanyproteinase,oftenreferredtoasapanproteaseinhibitor.InthebrainofAlzheimer"sdisease(AD)patients,α2Malsohasbeenlocalizedtodiffuseamyloidplaques,supportinganimportantroleforα2MinADetiopathology.Isabletoinhibitallfourclassesofproteinasesbyaunique"trapping"mechanism.Thisproteinhasapeptidestretch,calledthe"baitregion"whichcontainsspecificcleavagesitesfordifferentproteinases.Whenaproteinasecleavesthebaitregion,aconformationalchangeisinducedintheproteinwhichtrapstheproteinase.Theentrappedenzymeremainsactiveagainstlowmolecularweightsubstrates(activityagainsthighmolecularweightsubstratesisgreatlyreduced).Followingcleavageinthebaitregionathioesterbondishydrolyzedandmediatesthecovalentbindingoftheproteintotheproteinase.
