MultimericCD40L™isahighactivityconstructinwhichtwotrimericCD40ligandsareartificiallylinkedviathecollagendomainofACRP30.Thisconstructveryeffectivelysimulatesthenaturalmembrane-assistedaggregationofCD40Linvivo.Itprovidesasimpleandequallypotentalternativeto[CD40L+enhancer]combinations.MultimericCD40L™hasshowntosuppressalum-inducedIL-1βreleaseandcaspase-1activationinadose-,CD40-andtimedependentmanner,withoutaffectingBMDMviability.ItalsoeffectivelysuppressedtheinflammasomefunctiontriggeredbyNLRP3activators.Thesecretionofcaspase-1independentinflammatorymediatorshasbeenshowntobeunalteredorevenenhanced.
